An index file is the basic file that a webserver looks for if you haven't specified one. For example, when you go to http://google.com, you are actually accessing something like http://google.com/index.html -- here, index.html is your index file.
That message seems to indicate that you don't have an index.html or index.php file to default to. I would walk through the installation process. for joomla and phpnuke again. It seems that something went wrong during the install. Best of Luck, -Jess
